Your site content must be well written and structured. It does meant to be SEO optimised. All you need to use is proper paragraph structure, headings and sub heading, using images.
2. Site design:
You need to make sure your website is ready to run adsense ads. And, your wesbite should look good, simple is prefered. Site speed is secondary issue but matter the most. Even though adsense ads are A-sync but takes time to load. So, take care of that too.
Using other ads will not be the issue, unless you use spammy ad site like propeller.

Yes, as many of the blog authors suggested, the main aspect you need in this is, you must have quality contents on your page which directs people to land on your page via Google Bing and other search engines.
Along with the quality content, there are few more things you need to note, like the design, structure of your page etc.

Its not necessary to have 50 pages. Content should be unique.

First of all, you don't need any specific number of pages or posts to get the Adsense approval. All you need is quality content with some organic traffic.
Though not necessary, It would be easy to get the approval if you write at least 10 posts having more than 600 words. Also, the navigation of your blog plays an important role. Hence, choose a minimal theme with easy-access to all of your blog posts.
If you have already rejected, then wait for some time before applying again. Meanwhile, regularly update your blog with quality content and try to grab some organic traffic.
After 3-6 months of rejection, apply again and hopefully, you will get the approval.

Also Make Sure Your Pages Are "Indexed" On Google!,
At Applying For Adsense - Have A Domain Named Email, For example, If your Domain is "xyz.com", then make a gmail account named "email@example.com" & Apply For Adsense!

Here are Important Things For AdSense Approval:
- Try to have atleast 10 Posts Or More With Quality Content and not just copied from other websites.
- Try using adsense friendly theme like Magazine Pro in WordPress.
- Apply Only When You are Getting Organic Traffic and start getting some traction.
Adsense approval usually occurs within 24 to 48 hours now. If you create unique site that has never been involved with Google Adsense before, you should have no issues getting approved quickly. At that point, you will receive an approval email from Google that explains the process.
